A seventh U.S. Open title would break the professional era record for career grand slam titles that Williams now shares with Steffi Graf and leave her one behind all-time record holder Margaret Court. Bopanna, who partnered Gabriela Dabrowski in the mixed doubles, won the match after coming down from one set down to win 5-7, 6-3, 10-7 against Lukasz Kubot and Andrea Hlavackova.
In the singles arena, top male players Andy Murray, Stan Wawrinka and Kei Nishikori progressed into the next round of the competition.
The victory was her 308th at the four majors, breaking the record set by Federer, who is not competing at Flushing Meadows. She takes on seventh-seeded Italian Roberta Vinci – who shocked Serena en route to a runner-up finish past year.
Serena Williams, earned her 308th career major win, the most by any player in the Open era, by reaching the US Open quarterfinals.
After saving a match point in a five-set win in his last outing, Wawrinka was cruising against 63rd-ranked Illya Marchenko, who was playing in his first Grand Slam round of 16.
Williams, holding another service break in hand in the second set, soon served for the match just after the one hour mark.
Del Potro could face third-seeded Stan Wawrinka, who he defeated at Wimbledon, in the quarterfinals.
Pliskova, who had never before made it past the third round of a major in 17 prior attempts, saved a match point en route to a victory which scuppered a possible semi-final clash between Williams and her sister Serena.
Suarez Navarro, seeded 11th, was seeking the sixth Grand Slam quarterfinal of her career.
For Pliskova it was a breakthrough win, putting her into a first Grand Slam quarter-final.
Thiem took a medical timeout to have trainers look at his right knee after the second set’s fifth game.
Del Potro kept up his stunning form in recent months, with the Rio 2016 silver medallist knocking off 11th seed David Ferrer.
Del Potro had won the first set 6-3 and was up a break in the second.
The No. 2-seeded Murray, who is hoping to add Flushing Meadows to his Wimbledon and Olympic titles this year, takes on 22-seeded Grigor Dimitrov.
